The AS-SD1100 spore disruptor is a portable device to break open spores, pollen and other fine biological material suspended in liquid in standard Lab 2ml screw-top tubes (tubes with an outer diameter of 10.4mm). This has been found to release DNA, which can be used directly in diagnostic assays such as LAMP assays with minimal further processing such as a simple filtering step to remove coarse fragments. A further purification step is suggested for PCR assays. The sample requires addition of two plastic-coated ‘disrupting rods’ (Patent pending), which are also supplied by Agri Samplers. In addition, a minimum of 400 µL of liquid is needed to allow flow of the sample and movement of the disrupting rods, which are rotated by a magnetic field within the tube to disrupt the sample. The liquid used can include water, a buffer or LAMP assay mastermix. Run times of 4 minutes have been found to disrupt over 90% of wheat stem rust spores (Puccinia graminis tritici) and over 90% of Sclerotinia sclerotiorum spores. However it is suggested that users test an additional sample on first use and check by microscopy after different time points to ensure adequate disruption has been achieved before doing further work. The device is recharged using the power cable supplied connected to mains power.
